In vitro organogenesis and plantlet regeneration of ( Carica

چکیده
An efficient protocol was developed for micro-propagation of Indian papaya (Carica papaya L.) var. Co7. Shoot bud induction occurred on culturing epicotyl segments in MS (Murashige and Skoog, 1962) medium, followed by transfer onto shoot multiplication, elongation and rooting media. Adventitious buds were induced on MS medium supplemented with varying concentration of thidiazuron (TDZ: 0.5-10 μM). The highest shoot regeneration frequency and maximum number of shoots per explant was occured on MS medium amended with 2.5 μM TDZ. The explants with initiated shoot buds produced multiple shoots in MS medium supplemented with 5.0 μM 6-benzylaminopurine (BAP) and 0.05 μM naphthaleneacetic acid (NAA). Maximum shoot elongation was observed at 1.5 μM gibberlic acid (GA3). Upto 60.25% of the regenerated (elongated) shoots were rooted on MS medium containing 2.5 μM indole-3butyric acid (IBA) within 3 weeks after subculturing. The in vitro-raised plantlets were successfully hardened first under culture room conditions, then in greenhouse with 72% survival rate. The identified regeneration system could be efficiently used in various in vitro manipulation studies in papaya.

Plantlet Regeneration through Indirect Organogenesis of Flame Gold Tree (Koelreuteria elegans Laxm.)
Koelreuteria elegans, popularly known as “Flame Gold” is an ornamental tree. In vitro callus induction and regeneration from various explants (eaf segments and cotyledonary leaf) were studied on modified MS medium. The highest callus induction rate (80%) and multiplication was obtained in 2 mg/l 2,4-D from leaf segments. Calli transferred in 1.5 mg/l BAP resulted in efficient shoot regeneration...

Immature zygotic embryo explants of Carica papaya were cultured on MS medium supplemented with 2,4-D (2.0 mg/l) and formed globular embryos on explants without callus formation in 4-6 weeks. Maturation and conversion of somatic embryos was also achieved on the same medium. Cotyledonary stage embryos germinated to 63.66 and 68.33% in cv. honey dew and washington respectively in MS basal medium s...
متن کاملEffect of plant-derived smoke extract on in vitro plantlet regeneration from rapeseed (Brassica napus L. cv. Topas) microspore-derived embryos
The effect of aqueous smoke extract, derived from Tanacetum parthenium, onin vitro plantlet regeneration from rapeseed (Brassica napus L. cv. Topas)microspore-derived embryos (MDEs) was evaluated in this study. Inoculation ofrapeseed MDEs with the smoke-extract dilutions (1:250, 1:500, 1:1000 and 1:2000)for various times (5, 10 and 15 min) significantly enhanced the percentage ofregenerated pla...
متن کاملAn efficient protocol for in vitro regeneration of Podophyllum hexandrum, a critically endangered medicinal plant
Rhizome explants were used to develop a protocol for in vitro plantlet regeneration of Podophyllum hexandrum, a critically endangered medicinal plant, through direct organogenesis. Highest rate of multiple shoot formation was noted in MS medium supplemented with 11.42 μM IAA within three months. A synergistic effect of 2.68 μM NAA and 11.1 μM BAP was second best. The 1⁄2 MS liquid medium with 1...

An efficient protocol is suggested for in vitro culture of five Phalaenopsis hybrids obtained by hand-cross-pollination of three commercial hybrids Calgary, Ankara, and Kendall. Four nutrient media- namely half-strength Murashige and Skoog, Knudson, Phytamax containing activated charcoal, and Mitra– once supplemented (with coconut water, peptone, or both), and once without any supplement were c...
